Abstract

Les cèl·lules iNKT són una subpoblació de limfòcits T que mostren característiques pròpies tant de les cèl·lules del sistema immune innat com de l’adaptatiu, i per aquest motiu es considera que constitueixen un pont entre els dos sistemes. S’identifiquen per expressar un TCR semi-invariant (iTCR) que reconeix el glicolípid α-galactosilceramide (aGalCer) i altres antígens de naturalesa lipídica presentats per CD1d. Tot i la gran conservació estructural en el reconeixement del CD1d, la resposta de les iNKT contra un antigen determinat pot ser molt variable en funció de les característiques físico-químiques d’aquest, podent generar respostes quantitativa i qualitativament molt diferents amb una gran capacitat per modular l’activació de tot el sistema immune. Aquesta darrera característica ha fet que les cèl·lules iNKT siguin objecte d’estudi important per molts investigadors, que han centrat esforços en trobar reactius que permetin manipular l’activació de les iNKT i poder-los aplicar en el tractament de patologies en les que hi ha una implicació rellevant del sistema immune. En aquest treball s’ha estudiat l’activitat biològica i antitumoral d’un nou anàleg sintètic de l’αGalCer, anomenat AC11, amb una estructura més hidrofòbica i resistent a la degradació enzimàtica. Primerament s’ha descrit la seva capacitat per a ser reconegut i estimular a les cèl·lules iNKT tant de ratolí com d’humà in vitro. Després, utilitzant un model in vivo de ratolí s’ha descrit la capacitat de l’AC11 per induir una resposta de citocines i quimiocines totalment polaritzada cap a Th1, en contrast amb la resposta de tipus Th0 que indueix l’αGalCer. També s’han estudiat els efectes de l’administració in vivo d’aquest compost en l’ activació i expansió de diversos tipus de cèl·lules del sistema immune, en diferents òrgans i a diferents temps, veient que l’AC11 produeix una expansió més gran i prolongada que l’αGalCer de cèl·lules limfoides i mieloides. A més, s’ha comprovat la capacitat de l’AC11 per induir una forta resposta anti-tumoral contra el model de tumor hematològic transplantable de ratolí RMA, demostrant que aquest nou anàleg és més eficaç i potent que l’αGalCer, provocant una resposta quantitativa i qualitativament diferent. També s’ha estudiat la capacitat de l’AC11 per a actuar com a adjuvant en la generació d’una resposta específica antitumoral, tant en la vacunació contra un únic antigen proteic (ovoalbúmina) com en la vacunació conta múltiples antígens (cèl·lules necròtiques), veient que aquest agonista és capaç d’induir una resposta antitumoral específica. Finalment s’ha analitzat la viabilitat d’una nova teràpia que utilitza l’activació del sistema immune produïda per els agonistes iNKT per incrementar els efectes depleccionants d’un anticòs anti-CD20, i encara que per diversos motius no s’ha mostrat efectiu en el tractament d’un limfoma B, si que ha estat capaç d’augmentar l’acció eliminadora de l’anticós sobre les cèl·lules B.


The iNKT cells are a subpopulation of T lymphocytes that show characteristics of both the innate and the adaptive immune system, which is why they are considered to be a bridge between the two systems. They are identified by expressing a semi-invariant TCR (iTCR) that recognizes αGalactosilCeramida (aGalCer) and other lipid-derived antigens presented by CD1d. Despite the great structural conservation in the recognition of CD1d, the response of the iNKT cells to a given antigen can be very variable depending on the physico-chemical characteristics of the antigen, and can produce very different responses quantitative and qualitatively that have a great capacity to modulate the activation of the whole immune system. This last feature has made the iNKT cells an important object of study for many researchers, who have focused efforts on finding reagents that enable the activation and manipulation of iNKTs, to be applied to the treatment of pathologies in which there is a significant implication of the immune system. In this work, the biological and antitumor activities of a new synthetic analog of αGalCer, called AC11, with a more hydrophobic structure and resistant to enzymatic degradation, have been studied. Firstly, its ability to be recognized and stimulate murine and human iNKT cells in vitro has been described. Then, using an in vivo mouse model, the AC11 compound has been shown to induce a cytokine and chemokine response totally polarized to Th1, in contrast to the Th0 response that induces aGalCer. The effects of the in vivo administration of this compound on the activation and expansion of several immune cell types in different organs and at different times have also been studied, observing that the AC11 produces a larger and more prolonged expansion of lymphoid and myeloid cells than the αGalCer. In addition, the ability of AC11 to induce a strong anti-tumor response against the model of transplantable mouse tumor RMA has been proven, demonstrating that this new analog is more effective and potent than αGalCer, causing a response quantitative and qualitatively different. The ability of AC11 to act as an adjuvant in the generation of a specific antitumour response has also been studied, both in the vaccination against a soluble protein antigen (OVA) and in the vaccination with multiple antigens (necrotic cells), observing that this agonist is able to enhance a specific antitumor response. Finally, we analyze the feasibility of a new therapy that uses the activation of the immune system produced by the iNKT agonists to increase the depleting effects of an anti-CD20 antibody. Even if, due to various reasons, such effects have not been observed in the treatment of mouse a B cell lymphoma, the compound has been able to increase the action of the antibody in eliminating endogenous B cells. In conclusion, the AC11 compound has many characteristics that make it of great interest as a potential agent in anti-tumor treatments.
