Abstract

Summary Validamycin A (Val. A) has been used successfully in the fight against phytopathogenic fungi in crops. Here, its putative antifungal effect against the human pathogen Candida albicans is examined. Val. A acts as a potent non-competitive inhibitor of the cell wall-linked acid trehalase (Atc1p). The estimated MIC50 for the C. albicans parental strain CAI-4 was 0.5 mg/ml. A homozygous atc1Δ mutant lacking functional Atc1p activity showed greater resistance to the drug. However, the antifungal power of Val. A was limited compared with the lethal action of amphotericin B (AmB). The endogenous content of trehalose rose significantly with both drugs although no synergistic effect was observed between them. It seems therefore that inhibition of the trehalases is not lethal in C. albicans. Catalase activity was not affected by Val. A, so that this antioxidant activity does not play a major part in protecting the cell integrity in the case of treatment with Val. A. The drug did not impair the formation of germ-tubes, confirming that trehalose hydrolysis is not essential for morphogenesis. In addition, C. albicans cells and human macrophages showed a similar sensitivity to the compound. Therefore, Val. A cannot be considered a clinically useful antifungal against C. albicans. On the other hand, recent evidence suggests that the lethal effect induced by some clinical antifungals depends on previously unknown targets. In this study, we also examine the hypothetical role played by the intracellular formation of ROS in the fungicidal action of AmB and micafungin (MF) on C. albicans. The clinical MIC90 for MF and AMB were 0.016 and 0.12 mg/L, respectively (SC5314 strain), although CAI-4 cells showed a similar sensitivity to both drugs in YPD medium. The polyene AMB strongly induced ROS production in parallel with a severe degree of cell killing in both CAI-4 and SC5314 strains, whether grown in PBS or YPD. However, the fungicidal effect of MF was more effective in YPD, when it showed a higher degree of ROS generation and cell toxicity than in PBS. Preincubation with rotenone or thiourea reduced ROS production and caused a marked increase on cell viability, regardless of the antifungal applied. In contrast, AMB promoted higher intracellular synthesis of trehalose, which acts as a specific protector against oxidative stress in C. albicans. In C. albicans, the MAP-kinase Hog1 pathway plays an essential protective role against oxidative stress. The hog1Δ mutant showed a high degree of sensitivity to AmB, but no differences were observed with MF compared to its parental strain, RM-100. These results firmly support the involvement of Hog1 in the resistance of C. albicans to AmB, and point to the induction of an internal oxidative stress in C. albicans through the release of ROS as a contributory factor to the antifungal action of AMB but not MF. The simultaneous measurement of several antioxidant enzymes (catalase, glutathione reductase and superoxide dismutase) showed that these activities were strongly induced in cells treated with AmB and MF, such activation being especially pronounced in the hog1Δ mutant. We also investigated the in vitro correlation between the fungicidal effect of AMB and its hypothetical inhibitory action on hypha formation in C. albicans. The inhibitory effect of AmB on germ-tube formation was due to the high toxicity of AmB, whereas the regulatory elements involved in the dimorphic transition were not involved. Fungicidal concentrations of MF also supressed hypha formation and altered the morphology of C. albicans cells. In addition, MF increased the recognition and activation of macrophages of the innate immune system by increasing the production of the proinflammatory cytokine, TNF-α, and the anti-inflammatory, IL-10, that facilitate removal of the fungal infection.


Resumen La validamicina A (Val. A) se ha empleado satisfactoriamente para combatir hongos fitopatógenos que afectan a las cosechas. En esta Memoria hemos analizado su posible acción antifúngica contra Candida albicans. Val. A se comportó como un inhibidor no competitivo de la trehalasa ácida de pared (Atc1p), con una CMI50 de 0,5 mg/ml para la cepa parental CAI-4 de C. albicans. El mutante homocigótico nulo atc1Δ, carente de la actividad Atc1p, mostró una mayor resistencia al efecto de la droga. Sin embargo, el poder antifúngico de la Val. A fue limitado en comparación con anfotericina B (AmB), mucho más tóxica. El contenido intracelular de trehalosa aumentó significativamente al aplicar ambas drogas, aunque no se observó un efecto sinérgico entre ellas, por tanto, parece que la inhibición de las trehalasas no es letal para esta levadura. La actividad catalasa resultó alterada, por lo que no parece jugar un papel crucial en la defensa de la integridad celular tras el tratamiento con Val. A. Del mismo modo, este compuesto no afectó a la formación de tubos germinativos, confirmando que la hidrólisis de trehalosa no es esencial durante la morfogénisis. Además, los macrófagos humanos mostraron una sensibilidad a Val. A similar a la observada en C. albicans, lo que descarta su utilidad clínica. Por otra parte, estudios recientes indican que parte del efecto letal de ciertos antifúngicos clínicos se debe a su acción sobre dianas moleculares secundarias, aún desconocidas. En este estudio, hemos analizado la producción de ROS y su implicación en el efecto fungicida de AmB y micafungina (MF) sobre C. albicans. La CMI90 para MF y AmB fue de 0,016 y 0,12 µg/ml, respectivamente (cepa SC5314), mostrando la cepa CAI-4 un comportamiento similar en medio YPD. AmB indujo una fuerte producción de ROS junto a una elevada letalidad en células CAI-4 y SC5314, en YPD y PBS. Sin embargo, MF fue más letal en YPD y con una mayor producción de ROS que en PBS. No obstante, al preincubar las células con rotenona o tiourea, se redujo el nivel de ROS intracelular y se incrementó la viabilidad con ambas drogas. Además, AmB aumentó la síntesis de trehalosa endógena, que actúa como un protector frente a estrés oxidativo en C. albicans. La ruta de las MAP-Quinasas, Hog1, cumple un papel protector esencial contra estrés oxidativo en C. albicans. El mutante homocigótico nulo, hog1Δ, mostró una elevada sensibilidad a AmB respecto a la cepa parental RM-100, a diferencia de MF, cuya acción fue similar en ambas cepas. Estos resultados ponen de manifiesto la importancia de Hog1 en la resistencia de C. albicans a AmB, y el papel de ROS intracelular como factor implicado en la acción antifúngica de AmB, aunque no de MF. No obstante, los dos antifúngicos provocaron una fuerte inducción de las enzimas antioxidantes: catalasa, glutatión reductasa y superóxido dismutasa, especialmente en el mutante hog1Δ. También se analizó la correlación in vitro entre el efecto fungicida de AmB y su hipotética acción sobre la formación de hifas en C. albicans. El efecto inhibitorio producido por AmB sobre la transición dimórfica fue causado por su elevada toxicidad y no por su acción sobre componentes implicados en la morfogénesis. Por su parte, MF también inhibió la formación de hifas y alteró la morfología de C. albicans. Además, MF incrementó el reconocimiento y la activación de los macrófagos, manifestado mediante el incremento en la producción de la citoquina proinflamatoria TNF-α y la antiinflamatoria IL-10, lo que facilita la eliminación de la infección fúngica.
